Abstract:
To address accuracy challenges in polysaccharide methylation analysis, a library of partially methylated alditol acetates (PMAAs) was constructed. The chromatographic behavior of these standards was systematically characterized on three columns with different polarities: high-polarity (HP-INNOWax), mid-polarity (DB-23), and low-polarity (Rxi-5 ms). A dataset containing relative retention times (RRT), resolution (Rs), and epimer selectivity factors (α) was generated. Based on this, a standardized analytical workflow was proposed for the first time, using the PMAA separation behavior database as a reference, chromatographic separation as the core, and mass spectrometry as an auxiliary identification method. This workflow was validated through accurate glycosidic linkage analysis of pullulan and a heteropolysaccharide. This study contributes a comprehensive dataset and analytical workflow aimed at improving the accuracy and standardization of polysaccharide methylation analysis.
