Background:

  • Autism spectrum disorder (ASD) is linked to altered glial function and increased proinflammatory cytokines.
  • These changes are associated with behavioral and cognitive impairments in individuals with ASD.

Purpose of the Study:

  • To investigate neuroinflammation in ASD.
  • To explore the relationship between neuroinflammatory markers and autism symptom severity.

Main Methods:

  • Gene expression of NLRP-3 and RANK-RANKL-OPG inflammasome pathways was analyzed in children with ASD, siblings, and controls.
  • Severity of autism symptoms and neuro-psychiatric dysfunction was assessed using standardized scales.
  • Correlation analysis was performed between gene expression and clinical scores.

Main Results:

  • Up-regulation of inflammatory genes (IL-1β, Casp1, NLRP3, NLRP1, TNFRSF11B, TNFRSF11A, TNFSF11) was observed in the ASD group.
  • Statistically significant differences were found for TNFRSF11B, TNFRSF11A, and TNFSF11 between ASD cases and healthy controls.
  • Correlations were identified between inflammatory pathway genes and linguistic, cognitive, sensory, and neuromotor domains.

Conclusions:

  • This study provides the first evidence of RANK-RANKL-OPG pathway activation in ASD.
  • The findings highlight the detrimental impact of RANK-RANKL-OPG and NLRP3 inflammasome complexes on neurological development in ASD.
