Area of Science:

  • Neuroscience
  • Genetics
  • Imaging Technology

Background:

  • Genetically encoded calcium indicators like GCaMP are crucial for monitoring neuronal activity.
  • Existing GCaMP transgenic rat lines have limited genetic variation for diverse neuroscience needs.

Purpose of the Study:

  • To establish novel transgenic rat lines expressing GCaMP8 for enhanced neuronal activity monitoring.
  • To provide a new tool for functional brain mapping and cortical macro-imaging.

Main Methods:

  • Created two transgenic rat lines: one with Thy1 promoter-driven Cre recombinase and another with a floxed STOP cassette followed by GCaMP8 at the β-actin locus.
  • Mated these lines to produce Thy1-Cre-driven GCaMP8-expressing rats.

Main Results:

  • GCaMP8 expression was observed in approximately 80% of cortical neurons.
  • Transcranial calcium imaging in anesthetized rats detected responses to 68 dB click sounds (ΔF/F0 = 1.2%).
  • Peak response rise time and half decay time were both approximately 150 ms.

Conclusions:

  • The developed GCaMP8-expressing rats serve as a valuable tool for cortical macro-imaging.
  • These rats facilitate functional mapping using a simple transcranial approach.
  • The parental transgenic rat strains also offer utility for neuroscience studies.
