Association of the TGF-beta1 polymorphism with primary open-angle glaucoma: a case-control study

• Primary open-angle glaucoma (POAG) is linked to reduced aqueous humor outflow.

• The TGF-beta1 gene's -800 G>A polymorphism (rs1800468) was investigated for its role in POAG pathogenesis.

Purpose of the Study:

• To investigate the association between the TGF-beta1 gene polymorphism at position -800 G>A (rs1800468) and POAG.
• To determine if the TGF-beta1 -800 G>A polymorphism is a risk factor for POAG in patients from Khorasan Razavi Province, Iran.

Main Methods:

• A case-control study design was employed.
• 105 POAG patients and 105 age-matched healthy controls were recruited.
• DNA was extracted and genotyped for the TGF-beta1 -800 G>A polymorphism using PCR-RFLP.

Main Results:

• No significant differences in genotype frequencies (GG, GA, AA) were found between POAG cases and controls (P > 0.05).
• Allele frequencies (G vs. A) also showed no significant association with POAG (P > 0.05).
• Statistical analyses across various inheritance models (codominant, dominant, recessive, overdominant) revealed no significant associations.

Conclusions:

• The TGF-beta1 -800 G>A polymorphism does not appear to be significantly associated with the development of POAG in the studied Iranian population.
• Inheritance of the mutant A allele of TGF-beta1 at -800 is not identified as a risk factor for POAG in northeastern Iran.
