Measurement of T Cell Migration by T Cell-Dependent Bispecific Antibody in Cancer Cells and T Cell Cross-Linking

Abstract:
T cell-dependent bispecific antibodies (TDBs) have revolutionized the treatment of innovative cancer therapies with their potent anti-tumor effects, especially against hematologic malignancies. T cell responses to TDBs induce T cell migration, which is believed to play an important role in drug efficacy. Effective migration of T cells within the tumor microenvironment increases their contact with tumor cells and enhances T cell activation and cytotoxic activity against tumor cells. In this protocol, we present a method using TDB co-culture supernatants and a trans-well to evaluate T cell migration. This will provide a reliable approach to analyze in detail the mechanism of T cell migration in TDB-mediated immunotherapy and contribute to improving its drug efficacy.
