Terpenoids from Euphorbia ebracteolata (Hayata.) with their multidrug resistance reversal activity

Abstract:
One previously undescribed ursane-type triterpenoid euperata A (1) together with twelve known diterpenes, including one ent-isopimarane diterpene (2), one kaurane diterpene (3), and ten ingol diterpenes (4-13) were obtained from the Euphorbia ebracteolata Hayata. Their structures were established through extensive spectroscopic (e.g. NMR and HRESIMS). Besides, the absolute configuration of 1 was elucidated via ECD calculations. Furthermore, all the compounds were screened for their cytotoxicity and reverse multidrug resistance (MDR). As a result, compound 1 shows potential inhibitory activity against adriamycin-sensitive HepG-2 (Adr-sens HepG2), with IC50 value of 5.66 ± 1.15 μM. MDR reversal activity at a moderate level was shown by compounds 5, 8, and 10-11 in the HepG-2/Adr model, the reversal factors (RF) were 41.17, 86.23, 63.76 and 11.38, respectively.
