Abstract:
Here, we present a protocol to acquire, visualize, and quantify intracellular labile iron and oxygen signals at single-cell resolution using LiON (labile iron and oxygen notifier), a genetically encoded ratiometric reporter. We describe steps for fluorescence microscopy and flow cytometry acquisition, ratio-map visualization, and region of interest (ROI)-based quantification.
