The hypersensitive response (HR) represents a critical antiviral pathway in plants that can be leveraged for therapeutic screening via half-leaf spot assay. Attenuation of HR-induced necrotic lesions serves as a quantitative readout of antiviral efficacy. Here we present an integrated protocol combining: (1) rhizosphere bacteria isolation from asymptomatic plants in virus-infecting fields via serial dilution plating; and (2) quantitative antiviral assessment through necrotic lesion analysis. This comprehensive approach identifies microbial immunomodulators with potential for sustainable plant virus control.
