Area of Science:

  • Immunology
  • Cell Biology
  • Drug Discovery

Background:

  • Inflammasomes are key in immune responses and cellular signaling.
  • NLRP3 inflammasome activation is crucial for evaluating anti-inflammatory compounds.
  • ASC oligomerization, visualized as "specks" using ASC-GFP, indicates inflammasome activation.

Purpose of the Study:

  • To develop a high-throughput method for monitoring inflammasome activation.
  • To enable screening of potential anti-inflammatory compounds.
  • To provide real-time insights into inflammasome activation kinetics and toxicity.

Main Methods:

  • Utilized an Incucyte® live cell imager for real-time monitoring.
  • Employed ASC-GFP to visualize inflammasome speck formation.
  • Developed a methodology for higher throughput screening.

Main Results:

  • Demonstrated a platform for observing cell activation and death in real-time.
  • Enabled visualization from initial plating through inflammasome-induced cell death.
  • Provided insights into inflammasome activation kinetics.

Conclusions:

  • The developed methodology facilitates efficient inflammasome activation monitoring.
  • This platform is valuable for screening anti-inflammatory molecules.
  • Real-time imaging offers a comprehensive view of inflammasome activity and potential toxicities.
