Development and analytical validation of the first LAMP assays for the detection of Echinococcus vogeli

Abstract:
Neotropical echinococcosis, caused by Echinococcus vogeli, remains underdiagnosed in endemic regions due to the limited availability of accessible molecular tools. In this study, two loop-mediated isothermal amplification (LAMP) assays targeting the mitochondrial genes nad1 and cox1 were developed and analytically validated for the detection of E. vogeli, and were collectively designated as LAMPEV. Both assays successfully amplified E. vogeli DNA within 45-60 min. The LAMPEVnad1 assay, performed at 63 °C, showed a detection limit of 1 fg of genomic DNA, whereas the LAMPEVcox1 assay, performed at 58 °C, detected as little as 1 pg. No cross-amplification was observed with DNA from other parasites or host species, indicating high analytical specificity. The LAMPEVnad1 assay demonstrated higher analytical sensitivity, whereas LAMPEVcox1 showed amplification across a broader temperature range. These characteristics suggest distinct potential applications depending on laboratory conditions. Overall, the assays represent the first LAMP-based protocols for E. vogeli detection and provide a foundation for future applications in coprodiagnosis and epidemiological surveillance in endemic and resource-limited settings.
