A Study on the efficiency of protein extraction from oxytetracycline residues using the method of electrochemical

Abstract:
oxytetracycline residues were used as raw material to extract protein by electrochemical method. The initial pH, electrolyte dosage, voltage, plate spacing and moisture content (MC) were selected for single factor experiment, the response surface method was used to optimize the test parameters and explore the interaction between various factors. The principle of electrochemistry was explored by electron microscopy analysis and the experiment of free radicals quenching. The results showed that the optimum extraction conditions were as follows: initial pH = 7, electrolyte dosage = 5127 mg/L, plate spacing = 2 cm and MC = 95%; under these conditions, the protein extraction efficiency was 62.51%. The strength of effect of various factors on the protein extraction efficiency in the electrochemical reaction process was as follows: plate spacing > initial pH > electrolyte dosage > MC. The results of electron microscopy confirmed that the electrochemical oxidation method could destroy the mycelium cells of oxytetracycline residues and make the protein extraction. The experiment of free radicals quenching showed that the indirect oxidation played a major role in the electrochemical extraction of bacterial residue protein, and the hydroxyl radical oxidation played about 70% of the role in indirect oxidation. In addition, according to the cost estimation, it will cost about 403.64 ¥ to treat 1 t of oxytetracycline residues. The preliminary economic analysis showed that the electrochemical oxidation technology of protein extraction was economically favourable. The results of this study provide a reference for protein extraction from antibiotic residues.
