Background:

  • Rheumatoid arthritis (RA) is a chronic autoimmune disease causing joint inflammation and destruction, often affecting the knees.
  • Current treatments like intra-articular injections offer limited efficacy due to rapid drug clearance and poor targeting of neovascularization.

Purpose of the Study:

  • To develop and evaluate a dual-functioning embolic microsphere platform (ETN-GMA/MS) for sustained local delivery of etanercept (ETN).
  • To assess the therapeutic efficacy of ETN-GMA/MS in suppressing synovial inflammation and aberrant angiogenesis in a rabbit model of RA.

Main Methods:

  • Microspheres were fabricated using gelatin methacryloyl (GelMA) via microfluidic technology.
  • The platform combines genicular artery embolization with sustained release of etanercept (ETN).
