Abstract:
A ruthenium(II)-catalyzed remote alkylation of 2-pyridones has been developed via selective cleavage of either C-H or C-Cl bonds. Despite significant advances in ortho-C-H functionalization, distal modification of 2-pyridones remains challenging due to inherent regioselectivity issues and electronic bias. By leveraging a nitrogen-directed σ-activation strategy, this protocol enables C5-alkylation as well as C3,C5-dialkylation in the absence of a ligand. Notably, the method accommodates both C-H activation and directed C-Cl bond cleavage pathways, providing complementary reactivity patterns from readily accessible substrates. The transformation exhibits broad substrate scope, good functional group tolerance, and operational simplicity. This study expands the synthetic utility of ruthenium catalysis for remote functionalization of heterocycles and offers a practical approach for the late-stage diversification of biologically relevant 2-pyridone scaffolds.

Reduction of Alkynes to cis-Alkenes: Catalytic Hydrogenation
Like alkenes, alkynes can be reduced to alkanes in the presence of transition metal catalysts such as Pt, Pd, or Ni. The reaction involves two sequential syn additions of hydrogen via a cis-alkene intermediate.

Preparation of Alkynes: Alkylation Reaction
Alkylation of terminal alkynes with primary alkyl halides in the presence of a strong base like sodium amide is one of the common methods for the synthesis of longer carbon-chain alkynes. For example, treatment of 1-propyne with sodium amide followed by reaction with ethyl bromide yields 2-pentyne.
Alkynes to Aldehydes and Ketones: Hydroboration-Oxidation
One of the convenient methods for the preparation of aldehydes and ketones is via hydration of alkynes. Hydroboration-oxidation of alkynes is an indirect hydration reaction in which an alkyne is treated with borane followed by oxidation with alkaline peroxide to form an enol that rapidly converts into an aldehyde or a ketone. Terminal alkynes form aldehydes, whereas internal alkynes give ketones as the final product.
C–C Bond Cleavage: Retro-Aldol Reaction
In the first step, as depicted in Figure 1, the base deprotonates the β-hydroxy ketone at the hydroxyl group to form an alkoxide ion.
