    Intracutaneous immunization with Semliki Forest virus in mice induced delayed-type hypersensitivity without antibodies. This cell-mediated immunity, characterized by footpad swelling, could be transferred via lymph node cells.

    Background:

    • Delayed-type hypersensitivity (DTH) is a critical immune response.
    • Semliki Forest virus (SFV) is an important viral pathogen.

    Purpose of the Study:

    • To investigate the induction of DTH using inactivated Semliki Forest virus.
    • To characterize the immune response following intracutaneous immunization.

    Main Methods:

    • BALB/c mice were immunized intracutaneously with purified inactivated SFV.
    • Antibody levels and DTH reactions (footpad swelling) were assessed.
    • Cellular infiltrates were examined microscopically.
    • Adoptive transfer experiments using lymph node cells, spleen cells, and serum were performed.

    Main Results:

    • Intracutaneous immunization induced DTH without detectable serum antibodies.
    • Higher antigen doses via intracutaneous route failed to induce antibodies but elicited DTH.
    • Cyclophosphamide pretreatment enhanced the DTH response.
    • Microscopic analysis revealed mononuclear cell infiltration in the footpads.
    • DTH was transferable with lymph node cells, but not spleen cells or serum.

    Conclusions:

    • Intracutaneous immunization with inactivated SFV can induce a potent DTH response mediated by T-cells.
    • The study highlights a biphasic character of DTH induction and its cell-mediated nature.
