Objective:
To investigate the expression levels of suppressor of cytokine signaling 1 ( SOCS1) and 3 (SOCS3) in patients with myelodysplastic syndrome (MDS) and assess their clinical significance.
Methods:
A total of 74 MDS patients were included in our hospital from January 2020 to June 2022, and 56 patients with non-malignant hematological diseases were selected as control group. The mRNA expression levels of SOCS1 and SOCS3 in bone marrow mononuclear cells were measured using real-time quantitative PCR, and their relationship with clinical characteristics of MDS patients was analyzed to evaluate their value in assessing treatment outcomes and prognosis.
Results:
The mRNA expression levels of SOCS1 and SOCS3 in MDS patients were significantly lower than those in the control group (both P <0.01). There were certain differences in mRNA expression of SOCS1 and SOCS3 among different MDS subtypes, with patients in the MDS-EB group showing significantly lower expression than those in the MDS-SLD group (both P <0.05). Additionally, patients classified as relatively high-risk according to the IPSS-R prognosis risk stratification had significantly lower expression of SOCS1 and SOCS3 compared to the relatively low-risk group (both P <0.01). The patients with low expression of SOCS1 and SOCS3 had a lower overall survival rate, while with a higher proportion of progression to acute myeloid leukemia. The Cox regression analysis also indicated that SOCS1 has high predictive value for poor prognosis in MDS patients.
Conclusion:
Both SOCS1 and SOCS3 mRNA expression are significantly reduced in MDS and closely related to poor prognosis, which can be of great value for disease prediction and prognosis.
