Background:
Cancer metabolism has emerged as a critical area of study in cancer research. Dysregulated lipid metabolism has been observed in various cancer types, and this metabolic rewiring facilitates tumor growth, metastasis, and treatment resistance. Additionally, aberrant lipid metabolism is associated with poor patient prognosis.
Methods:
This review aims to provide a comprehensive overview of cancer metabolism, with a specific focus on lipid metabolism/lipogenesis and its association with resistance to third-generation epidermal growth factor receptor tyrosine kinase inhibitors (EGFR-TKIs).
Results:
Lipid metabolism/lipogenesis provides cancer cells with energy substrates and building blocks for membrane biogenesis. In parallel, resistance to third-generation EGFR-TKIs has emerged as a major challenge in the treatment of non-small cell lung cancer (NSCLC) driven by EGFR mutations.
Conclusion:
Understanding the intricate connections between lipid metabolism/lipogenesis and resistance to third-generation EGFR-TKIs is crucial for developing effective therapeutic strategies. Targeting lipid metabolism pathways holds promise as a novel therapeutic approach to overcome treatment resistance and improve patient prognosis. Further research is needed to better understand the underlying mechanisms and identify potential drug targets in lipid metabolism.
