Rabies01:28

Rabies

Rabies is a lethal zoonotic disease caused by a single-stranded, negative-sense RNA virus of the Lyssavirus genus, within the family Rhabdoviridae. Its primary mode of transmission to humans is through bites or saliva-contaminated scratches from infected mammals such as dogs, bats, raccoons, or foxes. Transmission can also occur if infectious saliva contacts abraded skin or intact mucous membranes, including the conjunctiva.Viral Entry and Early ReplicationOnce introduced at the bite or scratch...

Summary
This summary is machine-generated.

We developed a new method, single transcriptome assisted rabies tracing (START), to map neural connections. This technique identifies the specific cell types that connect to neurons in the mouse brain.

Area of Science:

  • Neuroscience
  • Molecular Biology
  • Genomics

Background:

  • Understanding neural circuits requires mapping connections at the cell-type level.
  • Current methods lack transcriptomic resolution for input mapping.

Purpose of the Study:

  • To present a novel protocol, single transcriptome assisted rabies tracing (START), for mapping neural inputs.
  • To combine monosynaptic rabies tracing with single-nucleus RNA sequencing.

Main Methods:

  • Developed START protocol for mouse cortex.
  • Included Cre-dependent helper virus injection and EnvA-pseudotyped rabies infection.
  • Detailed tissue microdissection, nuclei sorting, library preparation, and computational annotation.

Main Results:

  • Successfully identified transcriptomic cell types providing inputs to defined neuronal populations.
  • Provided a comprehensive protocol for researchers.

Conclusions:

  • START enables high-resolution mapping of neural connectivity.
  • Facilitates understanding of neural circuit function through transcriptomic cell-type identification.
