Abstract:
Tolfenamic acid-based prodrug was stabilized by disulfide bonds and loaded into a covalent organic framework (COF) with a high loading efficiency of 39.2 wt%. Under a weak acid tumor microenvironment, the prodrug was released and activated by glutathione (GSH). Singlet oxygen (1O2) and hydroxyl radicals (˙OH) were efficiently generated under ultrasound irradiation, and excellent antitumor efficacy was achieved by combined chemo-sonodynamic therapy.

However, cancer treatments can pose several challenges, as therapies used to kill cancer cells are generally also toxic to normal cells. Moreover, cancer cells mutate rapidly and can develop resistance to chemical agents or radiation therapy. Besides, all types of cancer cells may not respond to the same therapy.
