This study introduces a novel N-heterocyclic carbene (NHC)-catalyzed dynamic kinetic resolution (DKR) process for synthesizing chiral α-aryl α-amino acids. The efficient method provides access to valuable pharmaceutical precursors with high stereocontrol.
Area of Science:
- Organic Chemistry
- Asymmetric Synthesis
- Catalysis
Background:
- α-aryl α-amino acids are crucial building blocks in pharmaceuticals.
- Existing synthesis methods often lack efficiency or broad applicability.
Purpose of the Study:
- To develop a robust protocol for the asymmetric synthesis of unnatural α-aryl α-amino acids.
- To explore the utility of N-heterocyclic carbene (NHC)-catalyzed dynamic kinetic resolution (DKR).
Main Methods:
- Utilizing an NHC-catalyzed DKR process for α-aryl α-amino acid synthesis.
- Employing asymmetric synthesis techniques to achieve high enantioselectivity.
- Performing subsequent chemical transformations on the synthesized products.
Main Results:
- Achieved good yields and high enantioselectivities for a diverse range of chiral α-aryl α-amino acids.
- Demonstrated excellent functional group tolerance.
- Successfully transformed products into key precursors for bioactive molecules, ligands, and organocatalysts without enantioselectivity loss.
Conclusions:
- The NHC-catalyzed DKR protocol is an effective method for synthesizing valuable chiral α-aryl α-amino acids.
- Density functional theory (DFT) calculations confirmed the origin of high stereocontrol.
- The developed method offers a versatile platform for accessing complex chiral molecules.

Preparation of 1° Amines: Azide Synthesis
Azide ions act as good nucleophiles and react with unhindered alkyl halides to form alkyl azides. Alkyl azides do not participate in further nucleophilic substitution reactions, thereby eliminating the chances of polyalkylated products.
