We sincerely thank Alois Bonifacio and co-workers for their valuable comments on our work entitled "Three-Principal-Substrate SERS Profiling Enables Reliable Screening of Serum Biomarkers: A General Approach" (Anal. Chem. 2026, 98, 9273; DOI: 10.1021/acs.analchem.5c08093). The present response aims to address the concerns raised regarding the interpretation of SERS band assignments, as well as the suggestion that more rigorous validation of differential spectral features should be pursued in future studies.
