Sarcolemma-associated protein (Slmap) is crucial for sperm formation. Slmap knockdown causes severe defects in sperm development and triggers germ cell apoptosis in Drosophila melanogaster.

Area of Science:

  • Developmental Biology
  • Cell Biology
  • Genetics

Background:

  • Spermatogenesis is essential for male fertility, involving germ stem cell maintenance, meiosis, and spermiogenesis.
  • Previous work showed Sarcolemma-associated protein (Slmap) knockdown leads to male sterility.
  • The specific role of Slmap in spermiogenesis remained unclear.

Purpose of the Study:

  • To investigate the role of Slmap in Drosophila melanogaster spermiogenesis.
  • To identify the molecular mechanisms underlying Slmap-dependent sperm development.

Main Methods:

  • Slmap knockdown in early Drosophila germline.
  • Phenotypic analysis of spermiogenesis defects.
  • Transcriptomic analysis (RNA-seq).
  • KEGG pathway analysis and functional assays for apoptosis.

Main Results:

  • Slmap knockdown caused severe spermiogenesis defects: abnormal paracrystalline material, fused mitochondrial derivatives and spermatids, axonemal abnormalities.
  • Transcriptomics revealed altered expression of genes in cuticle development, plasma-membrane adhesion, and individualization complex.
  • Apoptosis pathways were activated, evidenced by increased TUNEL signals and altered Hid/Diap1 levels.

Conclusions:

  • Slmap is vital for maintaining structural integrity during spermiogenesis.
  • Loss of Slmap leads to morphogenetic failure and activates the Hid-Diap1 apoptotic pathway, resulting in male sterility.
