Abstract:
In this issue of Structure, Xue et al.1 present the cryo-EM structure of the Norrin-Tspan12 complex, revealing that Tspan12 directly binds a Norrin dimer without enhancing its affinity for FZD4. Instead, Tspan12 acts as a ligand organizer, enabling the assembly of a quaternary Norrin-FZD4-LRP5/6-Tspan12 signalosome. This study provides new insight into ligand-specific Wnt signaling.
