Cascade Aza-Piancatelli Rearrangement/Friedel-Crafts Alkylation: Stereoselective Synthesis of Cyclopenta[b]indole

An efficient cascade reaction has been developed for the stereoselective synthesis of functionalized cyclopenta[b]indole scaffolds using a sequence of aza-Piancatelli rearrangement followed by an intramolecular Friedel-Crafts alkylation reaction as a key step. This facile one-pot protocol is very general, exhibits excellent stereochemical control, and provides easy access to biologically relevant cyclopenta[b]indole scaffolds in very good yields.
