Mechanism of Compound Xinyi Nasal Drops in the Treatment of Allergic Rhinitis Through STAT3/IL-17A/NF-κB Signaling

Abstract:
Compound Xinyi nasal drops (CXND) is a traditional Chinese medicine nasal drop developed from an empirical clinical prescription for the treatment of allergic rhinitis and has been used for more than 20 years, with precise therapeutic efficacy. However, the mechanism of action of CXND in treating allergic rhinitis remains unclear. Investigating the impact of CXND on allergic rhinitis and further defining its potential active ingredients and mechanism of action were the objectives of this study. The effective components of CXND were determined by ultra-performance liquid chromatography, and the core genes and important pathways of CXND in the treatment of AR were analyzed by network pharmacology. Then, molecular docking verified component-target binding activity. The guinea pig model was induced by OVA to study the effects of CXND on the STAT3/IL-17A/NF-κB signaling pathway and related inflammatory factors. The fingerprint spectrum of CXND was established, and seven effective components were identified. It was found that the seven components might exert their therapeutic effects on allergic rhinitis through the STAT3/IL-17A/NF-κB signaling pathway. In vivo experiments have found that CXND can effectively reduce the levels of IL-6, IL-17A, and TNF-α while inhibiting the expression levels of STAT3, IL-17A, NF-κB p65 mRNA, and their phosphorylated proteins.
