Abstract:
Sudarshanam tablet, a polyherbal ayurvedic formulation, emerges as a particularly promising remedy with remarkable bioactive potential and are widely used in the treatment of various infectious fevers. The objective of the present study is to identify the unique compounds present in this tablet and to conduct the marker-based standardisation using HPTLC and HPLC analysis. LC-HRMS results showed the presence of andrographolide and its dimer in the methanolic extract of Sudarshanam tablet and its major ingredient plant, Andrographis paniculata. Also, in the aqueous extract of Sudarshanam tablet, kaempferol was found to be the major constituent. This tablet showed good antibacterial action against both gram-positive and gram-negative bacteria. Molecular docking studies revealed that andrographolide and kaempferol exhibited significant anti-inflammatory activity with a greater binding affinity. The anti-inflammatory potential of the tablet was confirmed by protein -denaturation inhibition assay. These results support the usage of this formulation in various fever and inflammatory conditions.
