Background:

  • Non-invasive prenatal testing (NIPT) relies on analyzing fetal material from maternal circulation.
  • Isolation of rare fetal cells is crucial for cell-based NIPT (cbNIPT).
  • Current methods for single-cell isolation can be time-consuming and lack high throughput.

Purpose of the Study:

  • To assess the efficacy of Fluorescence Activated Cell Sorting (FACS) for single-cell isolation of rare fetal cells.
  • To evaluate FACS as a method for cell-based non-invasive prenatal testing (cbNIPT).

Main Methods:

  • Blood samples (30 mL) were collected from 75 low-risk pregnant women (10-15 weeks gestation).
  • Fetal cells were enriched using magnetic activated cell sorting and stained.
  • Single fetal cells were sorted into PCR tubes via FACS and verified using Short Tandem Repeat (STR) analysis.

Main Results:

  • FACS sorted an average of 13.7 cells per sample.
  • STR analysis confirmed 8.2 fetal cells on average, representing 60.2% of sorted cells.
  • The procedure achieved an approximate 16-million-fold enrichment, with 1.3% of samples yielding no fetal cells.

Conclusions:

  • FACS is a viable automated method for single-cell isolation of rare fetal cells.
  • This technique offers faster, high-throughput cell isolation for cbNIPT.
  • Isolated fetal cells enable more comprehensive genetic analysis of the fetus.
