Fluorogenic fusion aptamer sensor design is complicated by troubleshooting with increased labor and cost. Secondary structure prediction software can help reduce the number of designs to screen experimentally. We propose using predicted equilibrium probability as an intentional design feature to create a Broccoli aptamer-based ADP sensor. This work demonstrates a more streamlined approach for fluorogenic fusion aptamer sensors by establishing landmarks to guide exploration of the sensor-transducer space.
