    Background:

    • Childhood trauma exposure (TE) is linked to heightened negative emotions, impaired cognitive control, and increased anxiety disorder risk.
    • Cognitive control relies on dorsal anterior cingulate cortex (dACC) glutamatergic (Glu) neurotransmission.
    • Investigating dynamic dACC Glu levels via ¹H functional magnetic resonance spectroscopy (¹H fMRS) can reveal impacts of emotional processing on cognitive control in youth with TE.

    Purpose of the Study:

    • To assess the impact of negative emotional processing on cognitive control mechanisms in adolescents with a history of trauma.
    • To investigate dynamic changes in dACC Glu levels using ¹H fMRS in relation to trauma exposure and emotional stimuli.

    Main Methods:

    • Fifty adolescents were divided into Higher and Lower childhood trauma exposure groups.
    • ¹H fMRS measured dACC Glu modulation during an inhibitory motor control task with emotional (Faces) and non-emotional (Squares) stimuli.
    • Task involved selective and non-selective response modes to assess cognitive control under different conditions.

    Main Results:

    • A significant interaction between trauma exposure group and stimuli condition was found (p=0.031).
    • Adolescents with higher TE showed significantly lower dACC Glu modulation during emotional (Faces) compared to non-emotional (Squares) stimuli (p=0.023).
    • Within the higher TE group, Glu levels were significantly reduced during emotional stimuli processing.

    Conclusions:

    • Trauma exposure group differences in adolescent dACC Glu modulation were evident during cognitive control with emotional stimuli.
    • ¹H fMRS is valuable for detecting trauma-related differences in task-related excitatory neurochemical dynamics.
    • Findings highlight neurochemical alterations in dACC associated with emotional processing and cognitive control deficits in adolescents with childhood trauma.
