Abstract:
The presence of an immunosuppressive tumor microenvironment in breast cancer impedes the effective delivery of inducible drugs to the tumor core and hinders immune cells from penetrating the vascular wall to enter the tumor, severely limiting the therapeutic efficacy of drugs that induce immunogenic cell death in cancer cells. In this study, we creatively designed a composite micelle (P2SM@Sun) featuring a terminal-charged "hydrophobic-hydrophilic-hydrophobic" tri-segmented structure. Combined with microneedle transdermal delivery technology, we developed a novel drug delivery system (PSD MN) capable of achieving efficient combined delivery of Sun and Dox at breast tumor sites for precise synergistic therapy. This system can achieve the combined delivery and precise controlled release of Dox and Sun at tumor sites. Among them, sunitinib enhances the ability of doxorubicin to induce immunogenic cell death, maximizes the release of DAMP, activates the systemic immune response, promotes the infiltration of CD8+ T cells, and stimulates the secretion of pro-inflammatory factors (TNF-α and IFN-γ), demonstrating significant synergistic anti-tumor activity. Overall, this work establishes a highly efficient and safe transdermal co-delivery platform for the treatment of breast cancer. By leveraging synergistic immune activation mechanisms, it offers broad application prospects for breast cancer therapy.
