Abstract:
Management of rice bakanae disease, caused by Fusarium fujikuroi, faces significant challenges arising from the development of fungicide resistance in the pathogen. Pydiflumetofen (Pyd) is a novel succinate dehydrogenase inhibitor (SDHI) and might offer potential in alleviating this crisis. In this study, the establishment of a sensitivity baseline revealed a unimodal distribution (EC50 = 0.0623 ± 0.0108 μg/ml), suggesting that the sensitive population is relatively concentrated. To assess the risk of resistance to Pyd in F. fujikuroi, two high-resistance mutants (PydHR), six moderate-resistance mutants (PydMR), and two low-resistance mutants (PydLR) were generated by fungicide mutagenesis. The PydHR mutants exhibited similarities in mycelial growth rate, sporulation capacity, germination rate of conidium, various osmotic stresses, and virulence, indicating that the Pyd resistant (PydR) mutants might form subpopulations with competitive advantages in the field. Analysis of nucleotide sequences of succinate dehydrogenase subunit-coding genes revealed three types of mutations in PydR isolates: FfSdhBH248Y + FfSdhC2A83V, FfSdhC2A83V, and FfSdhDD133G. The expression levels of FfSdhA/B/C1/C2/D in resistant double mutations RSS24-13 (FfSdhBH248Y and FfSdhC2A83V) all exhibited significant upregulation change; FfSdhC2 was dramatically upregulated by nearly 200-fold. These results indicate that point mutations and FfSdh gene upregulation expression may codetermine the Pyd resistance mechanisms in F. fujikuroi. Cross-resistance analysis revealed that the PydR isolates also exhibited resistance to SDHIs isofetamid and fluopyram, but there was no cross-resistance between benzovindiflupyr and fungicides from other classes. These results indicate that the resistance risk of Pyd is elevated to medium-high, and reasonable monitoring and management of resistance to Pyd should be carried out in F. fujikuroi.
Insights
Pydiflumetofen (Pyd) resistance in Fusarium fujikuroi, the cause of rice bakanae disease, is linked to specific gene mutations and increased gene expression. This suggests a medium-high risk, necessitating careful Pyd management strategies.
Area of Science:
- Plant Pathology
- Fungal Genetics
- Agricultural Chemistry
Background:
- Rice bakanae disease (RBD), caused by Fusarium fujikuroi, is a significant agricultural concern.
- Fungicide resistance in F. fujikuroi poses a major challenge to effective disease management.
- Pydiflumetofen (Pyd), a novel succinate dehydrogenase inhibitor (SDHI), shows potential for managing RBD.
Purpose of the Study:
- To establish a sensitivity baseline for Pyd in F. fujikuroi.
- To investigate the mechanisms of Pyd resistance in F. fujikuroi.
- To assess the risk of Pyd resistance and potential cross-resistance with other fungicides.
Main Methods:
- Fungicide mutagenesis was used to generate Pyd-resistant mutants (PydHR, PydMR, PydLR).
- Mycelial growth, sporulation, germination, osmotic stress tolerance, and virulence were assessed in resistant mutants.
- Nucleotide sequencing of succinate dehydrogenase subunit-coding genes (SdhB, SdhC2, SdhD) and gene expression analysis were performed.
Main Results:
- A unimodal sensitivity distribution for Pyd was observed (EC50 = 0.0623 ± 0.0108 μg/mL).
- Resistant mutants showed comparable fitness traits (growth, sporulation, virulence) to sensitive strains, indicating potential competitive advantages.
- Identified resistance-associated mutations include FfSdhBH248Y + FfSdhC2A83V, FfSdhC2A83V, and FfSdhDD133G.
- Significant upregulation of FfSdhA/B/C1/C2/D genes, particularly FfSdhC2 (nearly 200-fold), was observed in resistant isolates.
Conclusions:
- Point mutations in Sdh genes and their upregulated expression collectively contribute to Pyd resistance in F. fujikuroi.
- Pyd-resistant isolates exhibited cross-resistance to isofetamid and fluopyram but not benzovindiflupyr or other fungicide classes.
- The risk of Pyd resistance is assessed as medium-high, underscoring the need for vigilant monitoring and strategic resistance management.
