Detecting epidermal growth factor gene (EGFR) mutations in rare circulating tumor cells (CTCs) from lung cancer patients offers new therapeutic insights. This study demonstrates a method for capturing and analyzing CTCs for EGFR mutations using droplet digital PCR (ddPCR).

Area of Science:

  • Oncology
  • Molecular Biology
  • Genetics

Background:

  • Epidermal growth factor gene (EGFR) mutations are key for guiding lung cancer treatment.
  • Current methods for detecting EGFR mutations include tumor biopsies and circulating tumor DNA (ctDNA) analysis.
  • Analyzing circulating tumor cells (CTCs) for EGFR mutations can provide deeper insights into targeted therapy efficacy and patient prognosis, particularly in non-small cell lung cancer (NSCLC).

Purpose of the Study:

  • To present a protocol for capturing CTCs from whole blood.
  • To assess the molecular content of captured CTCs for EGFR mutations using droplet digital PCR (ddPCR).
  • To highlight the potential of this method for analyzing rare CTCs in NSCLC.

Main Methods:

  • Utilized a semi-automated, size-based cell isolation platform to capture CTCs from whole blood.

Employing Digital Droplet PCR to Detect BRAF V600E Mutations in Formalin-fixed Paraffin-embedded Reference Standard Cell Lines

Published on: October 8, 2015

  • Recovered captured CTCs for downstream molecular analysis.
  • Performed ddPCR with specific assays for EGFR pT790M and EGFR pL858R mutations on genomic DNA from captured cells.
  • Main Results:

    • Successfully enriched two lung cancer cell lines (A549 and H1975) from whole blood.
    • Accurately quantified EGFR mutations within the H1975 cell line using ddPCR.
    • Demonstrated the feasibility of capturing CTCs and analyzing their molecular content.

    Conclusions:

    • This protocol offers a viable approach for capturing CTCs and analyzing EGFR mutations.
    • The method has realistic potential for interrogating the genomic information of rare CTCs.
    • This technique can aid in personalized medicine for lung cancer patients by identifying actionable mutations.
