Chemical Synthesis and Amyloid β Aggregation Inhibition of Hippophaenin A as a Gluconic Acid-Containing Ellagitannin

Abstract:
Saji is valued for its highly nutritious berries, which are used in health foods, herbal medicine, and cosmetics. While its traditional uses are supported by previous studies, further research on its chemical components is needed to fully understand its therapeutic effects. As a component of Saji, hippophaenin A was isolated as an ellagitannin containing gluconic acid in 1991. This study developed a synthetic approach and investigated the biological activity of hippophaenin A. The synthetic method involved the conversion of the glucopyranose ring into an oxime and the subsequent formation of the gluconic acid moiety for the synthesis of hippophaenin A. An NMR comparison between the fully methylated synthetic and naturally occurring hippophaenin A validated the success of our total synthesis. The biological study found that hippophaenin A inhibits amyloid β aggregation. This study will contribute to further research on the chemical structures, reactions, and bioactivities of ellagitannins containing gluconic acid.
