Glofitamab in Heavily Pretreated Mantle Cell Lymphoma Patients: A Multicenter Real-World Case Series

Abstract

Background:

Relapsed or refractory mantle cell lymphoma (r/r MCL) remains difficult to treat despite therapeutic advances.

Methods:

This multicenter retrospective study included 10 patients with r/r MCL treated with glofitamab across five sites in Austria and Italy before European approval (January 2023-May 2024).

Results:

Patients were heavily pretreated (median 4.5 prior lines; 5/10 post-CAR-T). After a median of five cycles, responses were observed in 7/9 evaluable patients, including complete response in 4/9. Cytokine-release syndrome occurred in 7/10 (mostly Grade 1-2), including grade 3 events in 2/10, while ICANS occurred in 2/10 patients, including one Grade 4 event. Three patients received glofitamab as bridging therapy and all proceeded to CAR-T infusion.

Conclusion:

Glofitamab showed encouraging activity and manageable toxicity in heavily pretreated MCL, including as bridging therapy to CAR-T-cell treatment.
