Abstract:
Current research on the genetic diversity of Perca fluviatilis has been predominantly conducted in the Xinjiang region. To date, no cross-geographic comparative study has been reported between the native habitats in Xinjiang and the major aquaculture production areas in northern China. To elucidate the genetic background of different geographic populations of P. fluviatilis, this study analyzed eight distinct geographic populations using ten microsatellite markers. The results showed that the observed heterozygosity (Ho) of the ten markers ranged from 0.529 to 0.850, and the polymorphism information content (PIC) ranged from 0.630 to 0.884, all indicating high polymorphism (PIC > 0.5). The maximum genetic distance was 0.383 (Zhongwei population and Korla population), and the minimum genetic distance was 0.085 (Altay population and Wujiaqu population). UPGMA cluster analysis based on genetic distances, together with STRUCTURE analysis, divided the eight populations into three distinct clades. AMOVA revealed that 91% of the genetic variation resided within individuals. These findings indicate that the P. fluviatilis populations generally exhibit high genetic diversity and low levels of genetic differentiation. The Korla population is genetically more distant from the other populations, showing some degree of genetic differentiation and possessing a relatively unique genetic structure. This study identifies populations with high genetic diversity, thereby providing theoretical support for the establishment of core breeding populations and the selective improvement of new varieties of P. fluviatilis.
