Reserpine-induced hypotension through catecholamine and RAAS/nitric oxide/endothelin dysregulation

Objective:
This study aims to investigate the mechanisms underlying reserpine-induced hypotension.
Methods:
A rat model of hypotension was established by reserpine administration, followed by levodopa (L-DOPA) intervention. Arterial blood pressure, histopathologic changes in the heart and kidneys, the levels of norepinephrine (NE), the renin-angiotensin-aldosterone system (RAAS), and endothelial function markers were systematically evaluated.
Results:
Reserpine significantly lowered blood pressure and caused myofibrillar disarray, tubular epithelial degeneration, and elevated biomarkers of cardiac and renal injury. These pathologic changes were attributable primarily to depletion of NE and dysregulation of vasoactive regulatory systems. L-DOPA treatment restored NE and vasoactive mediator levels and markedly ameliorated hypotension and cardiorenal injury.
Conclusion:
Reserpine induces hypotension and cardiorenal injury through suppression of the catecholamine system, accompanied by dysregulation of the RAAS and nitric oxide/endothelin (NO/ET) system, whereas L-DOPA attenuates these pathologic changes.
