Peroxidasin (PXDN) expression changes in IgA nephropathy (IgAN) kidneys, showing reduced levels in proximal tubules and increased levels in fibrotic areas. This kidney fibrosis marker correlates with declining kidney function.
Area of Science:
- Nephrology
- Molecular Biology
- Pathology
Background:
- Peroxidasin (PXDN), a heme peroxidase, is involved in basement membrane assembly.
- PXDN is implicated in fibrotic processes, but its role in kidney disease is not well understood.
Purpose of the Study:
- To characterize PXDN expression in kidney biopsies from IgA nephropathy (IgAN) patients.
- To assess the association of PXDN with kidney fibrosis and function in IgAN.
Main Methods:
- Immunofluorescence staining of kidney biopsies to detect PXDN expression.
- Correlation analysis of PXDN levels with fibrosis markers (collagen III, collagen IV) and estimated glomerular filtration rate (eGFR).
Main Results:
- PXDN expression was reduced in proximal tubules and increased in the fibrotic interstitium of IgAN kidneys.
- PXDN correlated with collagen III deposition in glomeruli and interstitium.
- Higher PXDN expression in glomeruli, proximal tubules, and fibrotic interstitium was observed in patients with impaired kidney function (eGFR < 60 ml/min/1.73 m²).
- Glomerular PXDN inversely correlated with eGFR.
Conclusions:
- IgA nephropathy exhibits compartment-specific alterations in PXDN expression.
- PXDN is associated with interstitial fibrosis, collagen III deposition, and reduced kidney function in IgAN.
- PXDN may play a role in fibrotic remodeling and could be a therapeutic target for chronic kidney disease (CKD).
