This study details the synthesis of ubiquitin-7-amino-4-coumarinyl-acetic acid (Ub-ACA), a fluorogenic substrate for deubiquitinases (DUBs). The activated cysteine-directed protein ligation (ACPL) technique provides a high-yield method for producing pure Ub-ACA for enzymatic assays.
