Protocol for non-invasive genotyping of axolotls using skin swabbing

Abstract:
Fin clipping is the standard method for sampling axolotl DNA for genotyping, but it requires anesthesia and removing a piece of tissue. Here, we present a non-invasive protocol for axolotl DNA sampling and genotyping using skin swabbing. We describe the steps for extracting DNA from skin cells collected with cotton swabs and then detail how to sex genotype axolotls using PCR and gel electrophoresis. This protocol removes the need to cause an injury before regeneration experiments and improves axolotl welfare.
