Preparation of magnetic agarose microspheres by spray-chilling method and their application in IgG purification

Abstract:
In this study, magnetic agarose microspheres (MAM) were successfully prepared by spray-chilling method. The effects of process parameters such as spray height, aqueous phase flow rate and spray pressure on the morphology and particle size distribution of microspheres were systematically investigated, and the optimal preparation conditions (spray height of 130 cm, aqueous phase flow rate of 0.22 L/min, and spray pressure of 0.25 MPa) were determined. Spherical MAM with uniform particle size distribution was obtained. MAM was characterized by FT-IR, XRD, VSM, etc. The results confirmed that magnetic nanoparticles (MNPs) were successfully embedded in the interior of agarose, and MAM had good superparamagnetism and magnetic responsiveness. SPA-MAM was prepared by coupling MAM with recombinant protein A and used for the purification of IgG in rabbit serum. The results showed that the self-made SPA-MAM had good adsorption effect, and the maximum static adsorption capacity of IgG remained above 95% of the initial adsorption capacity after five adsorption-desorption cycles.
