A new matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) model accurately identifies Staphylococcus argenteus, distinguishing it from Staphylococcus aureus. This diagnostic tool improves clinical microbiology laboratory accuracy.
Area of Science:
- Microbiology
- Mass Spectrometry
- Genomics
Background:
- Staphylococcus argenteus is phylogenetically distinct from Staphylococcus aureus but difficult to differentiate using standard methods.
- Phenotypic and 16S rRNA gene sequencing show high similarity between S. argenteus and S. aureus.
- Current identification methods, including biochemical assays and MALDI-TOF MS, lack reliability in distinguishing these species.
Purpose of the Study:
- To develop and validate a MALDI-TOF MS-based model for accurate identification of S. argenteus.
- To establish a reliable diagnostic tool for clinical microbiology laboratories.
Main Methods:
- A multiplex PCR assay targeting crtM and NRPS genes was used as the reference standard.
- MALDI-TOF MS spectra from S. argenteus and methicillin-susceptible S. aureus (MSSA) isolates were analyzed using ClinProTools.
- A model was developed based on characteristic peaks and validated on independent isolate sets and clinical samples.
Main Results:
- Five characteristic peaks (m/z 5005, 5285, 5323, 6440, 6526) were identified for S. argenteus.
- A rule based on the presence of at least 4 of these 5 peaks achieved 100% specificity and sensitivity in development and validation.
- The model correctly classified all clinical isolates, outperforming conventional MALDI-TOF MS.
Conclusions:
- The developed MALDI-TOF MS identification model accurately distinguishes S. argenteus from MSSA.
- A simple peak-based rule provides a practical and effective diagnostic tool for clinical microbiology.
- This advancement enhances the ability to differentiate closely related staphylococcal species in clinical settings.
