    Area of Science:

    • Cell Biology
    • Pathology
    • Electron Microscopy

    Background:

    • Gaucher disease is a lysosomal storage disorder.
    • Gaucher cells are characteristic of the disease, accumulating in various tissues.
    • Their origin and precise ultrastructure are subjects of ongoing research.

    Purpose of the Study:

    • To investigate the ultrastructural surface morphology of Gaucher cells.
    • To compare surface characteristics across different Gaucher disease patients.
    • To correlate surface features with the proposed reticuloendothelial origin of Gaucher cells.

    Main Methods:

    • Examination of Gaucher cells from seven patients with Gaucher disease.
    • Utilized both transmission electron microscopy (TEM) and scanning electron microscopy (SEM).
    • Detailed analysis of cellular surface topography.

    Main Results:

    • TEM revealed no significant deviations from previously reported Gaucher cell ultrastructure.
    • SEM demonstrated considerable variation in surface structure, both between patients and within individuals.
    • Observed surface features included microvilli, ruffles, ridges, and blebs; phagocytosis was noted in two patients.

    Conclusions:

    • The diverse surface ultrastructure of Gaucher cells supports their relationship with the reticuloendothelial system.
    • Surface morphology is highly variable, highlighting cellular plasticity.
    • Further research into Gaucher cell heterogeneity is warranted.
