Anti-inflammatory phytochemicals from medicinal herbs based on senoinflammation schema for healthy aging

Abstract:
Aging is characterized by chronic inflammation involving a continuous, multifarious, low-grade inflammatory response that contributes to age-related chronic diseases. Recently, researchers have focused on age-related chronic inflammation and anti-inflammatory phytochemicals to ameliorate age-related degenerative conditions. As proposed earlier, senoinflammation is a broader view of age-related inflammation with an insightful perspective, which differs from the traditional phenomenal view. Recent advances have identified the senescence-associated secretory phenotype (SASP) as a central mediator of senoinflammation, driving chronic tissue inflammation and systemic aging. Here, we propose a conceptual synthesis and integrative framework linking senoinflammation with phytochemical-mediated modulation across cellular senescence, inflammatory signaling, and metabolic pathways, emphasizing SASP regulation, redox balance, and immune-metabolic crosstalk. Among the sixteen phytochemicals reviewed, curcumin and resveratrol exhibit the most extensive senotherapeutic activities, followed by ginsenosides, baicalin, myricetin, and other flavonoids, owing to their ability to target multiple senescence-associated signaling pathways and suppress the SASP; however, further studies are needed to establish their clinical efficacy and safety. We also highlight emerging evidence on organ-organ crosstalk in systemic aging and the differential anti-inflammatory effects of dietary interventions. A better understanding of the senoinflammation concept and phytochemical interventions will provide further insights and enable the development of more efficacious anti-inflammatory and anti-aging strategies, thus enhancing health and quality of life.
