Preparing a Pollen Reference Collection and Standardized Protocols for Aerobiological and Melissopalynological Pollen

Abstract:
Palynology, the study of pollen and spores, has developed from its early foundations in the seventeenth century into a multidisciplinary science with applications in taxonomy, environmental reconstruction, aerobiology, forensic investigations, melissopalynology, and archaeology. This chapter provides a comprehensive overview of methodological approaches central to both basic and applied palynology. The preparation and organization of the reference pollen collection (Palynotheca) are described as indispensable tools for identification, teaching, and comparative research. Detailed protocols for preparing fresh, acetolysed, and permanent reference slides are outlined, emphasizing their diagnostic value and long-term stability. Aerobiological monitoring is examined with particular attention to standardized volumetric sampling method, slide preparation, and data analysis for ecological and allergenic forecasting. Melissopalynological analysis is presented through its historical development and current applications in honey authentication, environmental monitoring, and biodiversity assessment, with protocols for the analysis of fresh and acetolysed honey samples as well as pollen loads. Finally, pollen germination tests are introduced as complementary techniques for evaluating viability and reproductive biology in agronomically relevant species. The integration of classical and applied methodologies demonstrates the central role of pollen analysis in advancing both scientific research and practical applications across diverse fields.
