Abstract:
Chromatin immunoprecipitation (ChIP) can selectively enrich chromatin associated with certain proteins, thus enabling observation of epigenetic alterations. With ChIP methods, various epigenetic signatures can be identified, allowing for the inference of transcriptional characteristics of chromatin regions. These biological features may lead to activation or inhibition of cancer-associated genes, thus underscoring the importance of ChIP studies in cancer research. ChIP from tissues can be more complicated than using cells grown in culture, and needs some additional optimization. We describe here the process of ChIP using mouse lung tissues (including tumors), and the DNA thus isolated can be further analysed using sequencing methods.
