UNITE® Technology: Translating lysosomal targeting vaccines for allergy therapy

Abstract:
Allergic diseases affect more than 30-40% of the global population and continue to rise in prevalence, creating a substantial clinical and economic burden. Current treatments-including antihistamines, corticosteroids, biologics, and allergen immunotherapy-primarily provide symptomatic relief or require prolonged treatment with variable efficacy and risk of adverse reactions. There remains a critical need for therapeutic strategies that can safely and durably reprogram the underlying immune response. UNITE® (Universal Intracellular Targeted Expression) Technology, developed by Immunomic Therapeutics, Inc., is a proprietary lysosomal-targeting vaccine platform designed to address this need by enhancing antigen presentation through the lysosome-associated membrane protein-1 (LAMP-1) pathway. By directing antigens to endolysosomal compartments, UNITE® vaccines promote efficient engagement of major histocompatibility complex class II (MHC II) machinery and support balanced immune modulation. In preclinical mouse models of allergic disease, UNITE® constructs encoding major allergens have demonstrated reduced basophil activation, lower allergen-specific IgE titers, and enhanced Th1-biased responses characterized by interferon-γ (IFN-γ) production and IgG4 induction. The platform's modular and expandable design enables rapid adaptation to diverse allergens and supports both DNA and RNA vaccine formats, facilitating integration across therapeutic areas including oncology, infectious diseases, and allergy. This versatility allows efficient customization of antigen payloads while maintaining consistent lysosomal targeting and immune-balancing properties. Collectively, these findings highlight UNITE® Technology as a promising next-generation approach for allergic disorders-capable of delivering safer, more durable, and more precisely targeted immune re-education than current standard-of-care therapies.
