Study of Unconventional Cytoskeleton Components Regulating Actin Contraction

Abstract:
The cytoskeleton, as the internal structural framework of the cell, is not only critical for maintaining cell shape but also plays a central role in regulating cell motility and other various cellular functions. Recent studies have shown that certain organisms rely on unconventional cytoskeletal components to carry out their unique biological processes. For example, studies on the single-celled eukaryotic predator Lacrymaria have revealed that this organism employs unconventional cytoskeletal structures to drive the rapid contraction of its "neck". In this study, we developed an integrated approach combining mass spectrometry (MS), immunofluorescence (IF), super-resolution imaging, subcellular structural analysis, and three-dimensional (3D) reconstruction. This methodology is not only well-suited for investigating Lacrymaria but also broadly applicable for exploring the presence and biological functions of unconventional cytoskeletal components in other eukaryotic systems.
