Abstract:
Immunolocalization relies on the sequential binding of a specific primary antibody to its target antigen, followed by detection with a fluorophore-conjugated secondary antibody to visualize the subcellular localization of proteins. This technique enables precise and high-resolution mapping of protein localization and distribution patterns after secretion. This chapter presents a detailed protocol for the immunolocalization of secreted proteins from Ustilago maydis, both in vitro and in infected maize leaves.
