Abstract:
Proteins perform essential structural, enzymatic, and regulatory functions, many of which depend on their subcellular localization. In plant-pathogen interactions, they are crucial in establishing virulence. The phytopathogenic fungus Ustilago maydis employs protein-based strategies, including effector secretion, cell wall remodeling, membrane-associated transport, and regulatory signaling mechanisms to infect the maize plant. To facilitate the study of these proteins, a set of optimized and reproducible protocols has been developed for isolating cytosolic, membrane-bound, secreted, and glycosylated protein fractions. These methods combine cryogenic disruption, specific lysis buffers, TCA-DOC precipitation, and affinity enrichment, providing a robust workflow for comprehensive protein analysis in fungal pathogenesis.
