Abstract:
Advances in biophysical and structural methods have made previously intractable human proteins and protein complexes viable targets for high-resolution structure-function studies and biophysical characterization. The production of complex human membrane proteins of sufficient quality and quantity, however, remains a key bottleneck. Here, we present a primer on the scalable and cost-effective production of membrane proteins in HEK293 cells, touching upon contemporary methodology within this general framework and offering examples of human ABC transporters successfully targeted in our lab using these pipelines.
