Abstract:
Quantitative real-time PCR (RT-qPCR) is a powerful and precise tool for analyzing gene expression, particularly suited for studying the embryonic transcriptome. Its high sensitivity enables the detection of low-abundance nucleic acids, which are essential for examining gene expression in early-stage embryos, where cellular diversity and rapid transcriptional changes pose unique challenges. This chapter provides a comprehensive guide to designing and conducting RT-qPCR experiments for preimplantation embryos, with a focus on accurate quantification of messenger RNA (mRNA) levels. The technique's extensive dynamic range, sequence specificity, and reliability make it invaluable for comparing mRNA levels across diverse biological samples and for validating data from other molecular approaches, such as microarrays or next-generation sequencing. This chapter outlines essential steps and considerations for obtaining accurate, reproducible gene expression data in embryonic research.
